Ritonavir and the path that seemed to disappear

Ritonavir stayed the same chemical compound. A different crystal arrangement changed which form became easy to obtain—and changed what the capsule could reliably do.

In plain words

A molecule can pack into more than one crystal structure without becoming a different chemical. Development work had found only the form later called Form I. In early 1998, capsule lots failed dissolution and investigators identified a previously unknown Form II. Under the cited conditions, Form II was more stable and substantially less soluble.

Once Form-II crystals existed, their surfaces could make later Form-II growth easier under suitable conditions. That did not prove what caused the first historical nucleus. The old route became hard to reproduce; the old form did not leave reality. Later controlled crystallization and milling studies produced Form I again.

Not observed is not impossible. Not reproduced is not permanently absent. A changed result can correct the map without changing the molecule.

A related cyclic-carbamate degradant nucleated Form II in an experiment and was proposed as a possible trigger. That shows a plausible route; it does not prove the cause of the 1998 event. Stories about a particular person or object carrying crystals between sites are unsettled too.
